¡¡¡¡Develop A Direct Solid Phase Lanthane Timeª²resolved Fluoroimmunoassay Reagent of PAPPª²Autilizing PVA

¡¡¡¡Abstract :Objective To develop a direct solid phase lanthane timeª²resolved fluoroimmunoassayreagent of PAPPª²A by synthesizing a complex of (SA)zª²(Bio)xª²PVAª²(BCPDA)yª²Eu3+. Methods  Polyvinylamine (PVA) was labeled with biotinª²streptavidin and europium chelate of 4,7ª²bis(chlorosulfophenyl)ª²1,10ª²phenanthrolineª²2,9ª²dicarboxylicª²acid(BCPDA). Using labeled PVA complex,We designed twoª²site sandwich timeª²resolved fluoroimmunoassay of PAPPª²A and evaluated assay characteristics of PAPPª²A kit .Results We successed synthesizing a conjuagete of (SA)zª²(Bio)xª²PVAª²(BCPDA)yª²Eu3+ and a reagent of PAPPª²A. Detection limits achieved were 0.7mIU/L.The calibration curve was linear 0ª²10000mIU/L.Intraª² and interassay imprecision (CV) was 3.89%ª²4.96% and 7.35%ª²9.27%.Recovery was 95.8%ª²104.2%. Conclusions The conjugate of (SA)zª²(Bio)xª²PVAª²(BCPDA)yª²Eu3+ synthesizied was reactive ,highly fouorescent. A sorts of kit could be synthesized ,taking advantage of it. The reagent of PAPPª²A was potentially suited for use in clinical with highly analytical sensitivity.
